yes you can boot from hard drive as long as the intel raid controller is set to IDE mode. if Intel raid controller is set to RAID or AHCI then you cannot boot from SAS.
you would want to use the 8 pin connector. as for the esata there is only one on that board which is on the rear i/o panel. it is the red one right below the 1394 port. do not use the two sata port that is perpendicular to the motherboard as that is for SATA connection and esata will not fit...
because p43 chipset came from p45. its just a tweaked version of the p45. main differences are p45 chipsets also allows ddr3 memory and support of two pciE x16. so to answer your question, p43 came from p45 and have those two difference thats why they both show up in cpuz as p45 chipsets.

you can use any of the red ports on there. do not use the orange and white because those are for drivexpert. as long as you use the red port then everything should be okay. primary or secondary doesn't really matter since you are using sata and have only one hard drive.

i've noticed that you mention that you want a board with decent overclocking. for that reason, i would go with the p5q pro board with the better Q9550 cpu. i have that same setup and overclocking is a breeze.

check the ASUS memory qvl for your board to see if its compatible with four sticks of ram. You might also want to set the recommended manufacturer timings and voltages in the bios. sometimes you might have to up the voltages a bit to get the board to post.
