Thanks, I do have it defined in smb.conf. My question was whether defining the share in smb.conf actually mounts the share, or whether it has to be mounted in a separate (prior) operation for smb.conf to be able to see it. The partition is empty, so based on the advice above, I'll just start...

Tivos use an ATA 100 interface, so there is a limit of 137 GB/drive. You can use larger drives, but only the first 137 GB will be usable. Most people use 2x120 GB, which is the largest configuration you can use without wasting drive space.
