Nightshift is pretty much like dayshift. You still work 8 hours a day, then go home. In my experience, nightshift does tend to get the crap work though that the other shifts don't want to bother with. Nightshift also tends to be a bit "rougher around the edges," so to speak. Other than that, the main difference is that you're working when the sun is down, which, if you're in a building with artificial lighting, as most have, doesn't really matter much.

But, it does have benefits: no 7am rush hour traffic, except when you're leaving work perhaps, and then you don't need to be in a rush to get somewhere on time, since you're just going home. And there is the shift differential at some businesses.

As far as getting used to it? No. You don't. You never do. There are guys there that have worked that shift for 20 years that aren't used to it.
Depends. I worked at Walmart for 2.5yrs nightshift. I was used to it after the first week. Then I went to college for a year, and now I'm back on nightshift working at a warehouse. It only takes me about 3 days to fully adapt my internal clock to the new pattern. I sort of brute-force it, staying awake a LOT longer than I normally would. For instance, if I usually would go to bed at 11pm and wake at 7am (college schedule), then instead of going to bed at 11, I'd stay awake until maybe 7am or something crazy like that, just putzing around on the computer. Then the next day, stretch it out until my nightshift bedtime of 1pm. Then it's the third day, and I'm adjusted to nightshift.

Doing that "swing shift" thing, I think it's called, when you're switching shifts constantly, that I could not handle. I need consistency, and some degree of predictability.

So I guess it's an individual thing. Some people don't mind it, others can't stand it. I can operate just fine regardless of what Earth's position relative to the sun happens to be.
