There isn't really a reason. Barely anything in Ms. Rowling's books are consistent or logical. The "R.A.B." nonsense is a great example of this.

Snape is a "half blood", which means one of his parents is muggle born and the other is of the magical world. His middle name is "Prince".

R.A.B. is Sirius' brother or something.

And he decided to refer to himself in this way in exactly one book because...? I know the reason given by the author, it's just that the reason sucks.

Regulus Black isn't the R.A.B. in question.

Edit: Wait, apparently it is. I thought it was the rat guy. Guess it's been a while since I read the books.

And yes, Regulus Black (forgot middle name) is R.A.B.

He was on the run from Voldemort and Death Eaters because he went traitor. He didn't destroy it, he merely hid it in time (well, he died in attempt, but house elf Kreacher, who helped him get to the cavern/lake, hid it at Black family home). I don't think he had the time nor the appropriate weapon/material to destroy the horcrux (locket).
