The IS250 will have a 6-speed manual option. Unfortunately, the IS350 will not. However, Lexus originally offered the IS300 in auto only, and then added in a manual a year or two later. It might be possible that the IS350 will also follow this route.

Yes, the Lexus IS350 is suppose to compete with BMW 3-Series. Very nice looking car. Much better the the current BMW 3 Series.
