Summarized transcript from the D3 gameplay panel:

-Potions are still in the game but not as core an element
I wonder if you can still put them on your hotbar? Might move to system like WoW where pots are on cooldown, who knows.
-Says the story isn't going to be forced in front of your face
Likely a reaction to how D2 evolved over the years. People liked the story initially but later they just want to fight monsters and have fun, not sit through cut scenes/dialog.
-When questioned about group size, Blizz stated "once you get more than four or five people onscreen, things get kind of chaotic"
Possible "soft cap" of 4-5 (most effective groups at that size) but probably same or larger player cap then D2 is my guess
-No grid inventory, but no final inv size settled upon
You can see this in the gameplay video, each item takes up one box. Not sure how i like this. As people have pointed out in other forums, some of the charm is seeing the full item graphic. Hopefully they'll still have the art appear when you hover over the items
-They haven't figured out how many classes will be in yet
Looks like the 5 total classes stated on the D3 page isn't final. More speculation on this later
-Dev comments indicate possibility of respeccing skills
A very nice change from a character freedom perspective, wonder if that will hurt the longevity at all. No need to replay earlier levels with the same class.
-Witch doctor is not the evolved version of D2's necro
Does this mean the necro is one of the classes? Who knows.
-No items that drop for groups only (ie there's no WoW style raids), all items that drop for groups can drop for single players
Good, glad they're keeping important gameplay aspects separate from other genres/franchises
-Does not require DX10, should be very scalable to a range of systems
If that's the case I imagine it should run perfectly fine on modern day hardware
-Difficult to support mods/map editors
This probably means mods will be unlikely or only unofficially supported
