Not sure if it's been posted in this thread yet, but if your game slowness (FPS not lag) is happening during Maghda (act II), it might be caused by your antivirus software. Microsoft Security Essentials realtime protection goes nuts during this encounter. It must see something in the executable it doesn't like.
Blizz knows about it, but hasn't fixed it yet. There are a couple of other av software out there that do the same thing.
To fix it in MSE, I just added Diablo III.exe to the process exclusion and it's fine now. You could also just turn off realtime scanning during this boss battle.
