Sealing a box

I am sealing a compartment to make a speaker cabinent.

The edges of the compartment are about the width of a finger in distance.

Now, if I wanted to seal it, could I use that "Tuff Stuff" house foam? That stuff will seal a crack and gets really hard and airtight.

Also, is a port about the size of a teenager's closed fist too big? It's pretty long too. It's not really a port, but a long tunnel that runs along the side, and I am hoping it may just act as one. Hmm, maybe I can seal that too if I have to...

correct me if I'm wrong, but there is a formula to determine the size of a port hole, something having to do with speaker size, size of box, blah blah, I'm sorry I dont know it..

As for sealing, when I've built boxes I've always ran a bead of liquid nails on the edge of the wood and then screwed the two peices together. You can probably use liquid nails and still run it around the edges of the inside of the box, otherwise I would use a silicon or something similar. I'm pretty sure that the foam insulation wouldn't be good for sealing, beucase there are little holes all over the place.
