It takes a *VERY* picky ear and a likely an incredibly high end speaker/system to notice...and even then it's probably more mind over matter. I could see it happening with some bright tweeters more than anything else.

A $150 pair of Polk R50's probably don't count
Polk would have done a better job if they ended the grill just under the bottom driver and didn't do the wrap around the port.

Meh, they always sounded very different to me.

And actually the better speakers are designed to take into account the grill, it's the non high end speakers that do best without the grills. Try it and see.

If you can close your eyes and hear when somebody walks in front of you from how the soundstage changes then you can tell the difference between grill or not.

1, but you don't need the center channel for that setup.

A thin layer of fabric 1" away from the drivers basically does nothing to the sound.

Leave the grills on and move those main speakers out about 2' away from the side of that stand...they are too close together.
