Would probably get better responses if you changed the question to "presuming Hillary Clinton gets elected, what do you think her #1 priority should be in the beginning of her term"? Forget the 100 days since that's an arbitrary number and realize that most Presidents only have enough political capital for one major initiative, in Obama's case it was Obamacare - I know some will argue for the stimulus too but that was really a continuation of the Bush stimulus measures.

Maybe. Maybe not. Obama has turned immigration 180 degrees without any input or cooperation from Congress. Hillary will be in at least as strong a position. If the Dems manage to take the Senate too, then by the end of her first term we'll probably be seeing SCOTUS mandates for a lot of the far left ideas, like community standards, the Fairness Doctrine, and imputed income taxation.Depends on turnout and down ballot results.
She will enjoy a strong popular and electoral college lead, but that doesn't necessarily translate into a mandate, especially if turnout is historically low.
If the Dems don't take the Senate, she is essentially politically dead on arrival.

Beyond that, the house impeachment machinery will be getting warmed up from November to January ready for her to take office. The next four to eight years will be a total shitshow so long as the GOP holds both houses. See the Benghazi hearings as an example of what to expect stuck on a loop for her entire term(s).
With a Senate majority (or at least a tie), I forsee her getting a SCOTUS judge or two. However, an even bigger issue is that of the judiciary as a whole. Republicans have been dragging their feet for many open federal judge-ships for the past eight years, to the point where a continuation for another four or eight years would put the entire federal court system in jeopardy. I also forsee some effective foreign policy with regards to treaties, etc. moving forward as the House isn't involved in that. Other than that, she will face the same deadlock that hampered Obama for budget issues.
With Democratic control of both houses, I forsee new scotus judges to turn the court to its pre-Thomas/Scalia balance. Obamacare could finally be tweaked to fix its issues, including a public option. The floodgates of progress will finally be unleashed for two years, and she will need to coast on that for another six years like Obama did. GOPers won't let that last long.
