Check out the band Metric.
That is Emily Haines. I do not care for Metric very much (they're okay), but her recent solo album, "Knives Don't Have Your Back" (as Emily Haines & The Soft Skeleton), is a solid debut, especially "Doctor Blind" (the single) and "Winning" (my favorite track). Her style is completely opposite of her performances with Metric. Songs structure is more Elliott Smith doing piano ballads than sweaty rockers. "Winning" and a liberal pour of Maker's Mark over ice is my current favorite method of decompression. I have a copy where she sings "but they do" at the key line that completely twists the song. Love it.

Dresden Dolls, Amanda Palmer writes, sings, and plays the piano for all their songs.
The Fiery Furnaces, A true brother sister duo band, Eleanor Friedburger does the majority of the vocals and also writes some of the songs.
Benton Harbor Blues is flipping wonderful. One of the best songs of 2006. The Furnaces are a bit eccentric though.

Nina Perrson (Caridgans) is highly underrated. She is also smoking hot.

Her voice is drugged sex.

I like Leslie's solo album, but "7/4 (Shoreline)" by Broken Social Scene is my favorite thing she's ever done. It rocks my ---- off.
Replying to this post is really hard because I am a sucker for a good female lead vocalist. Any list I do will inevitably leave off a dozen or more favorites. I'll try for some that weren't mentioned (no particular order):
Harriett Wheeler (The Sundays)
Nina Simone
Amy Milan (Stars, and other bands)
Tanya Donnelly (Throwing Muses, Belly, solo)
Regine Chassagne (Arcade Fire - "in the backseat" especially)
Halfdis Huld (She did a vocal for an FC Kahuna track called "Hayling" that I love. Ultra smooth)
Emma Anderson & Miki Berenyi (Lush)
Bilinda Butcher (My Bloody Valentine - especially "Only Shallow")
Toni Halliday (Curve)
Edith Piaf
Jenni Lewis (Rilo Kiley)
... gotta get back to work, more later.
